User Guide: Method Transfer Calculator

Use this tool to geometrically scale analytical methods from conventional HPLC to UPLC (or vice versa), maintaining separation quality while optimizing speed.

1. Input Source Method Parameters

Enter the column details ($L$, $dc$, $dp$), Flow Rate, and Injection Volume of your current validated method.

2. Input Target Method Parameters

Enter the dimensions of the new column (e.g., UPLC 1.7 µm column). The calculator will instantly preview the scaled Flow Rate and Injection Volume.

3. Dwell Volume (System Delay)

Enter the Dwell Volume ($V_d$) for both systems if known. This allows the calculator to adjust the gradient timing to ensure the solvent composition reaches the column at the correct relative time.

4. Gradient Table

Enter your original gradient time program and composition (%A, %B, %C). Clicking Transfer Method will calculate the new time points for the target column.

Key Scaling Formulas (USP <621>)

F₂ = F₁ × (dc₂/dc₁)² × (dp₁/dp₂)
V₂ = V₁ × (L₂ × dc₂²) / (L₁ × dc₁²)
t₂ = t₁ × (V₀₂ / F₂) / (V₀₁ / F₁)
